Distribution and co-localization of choline acetyltransferase and p75 neurotrophin receptors in the sheep basal forebrain: implications for the use of a specific cholinergic immunotoxin

Experimental procedures
Animal care and all procedures were in accordance with authorization A 37801 of the French Ministry of Agriculture. All efforts were made to minimize both the suffering and the number of animals used.
Choline acetyltransferase labeling
The peroxidase staining appeared to be homogeneous in the perikarya and the proximal dendrites (Fig. 2a, c, e). The nucleus was never stained. No difference in labeling intensity was observed between ewes. No immunoreactive neurons were observed on sections preincubated with an excess of ChAT.
Discussion
The results of the present study provide evidence in sheep concerning: (i) cholinergic neuron distribution in the basal forebrain (MS, VDB, HDB and NBM) and in the striatum, using ChAT immunohistochemistry, (ii) the co-localization of ChAT and p75NTR labeling in the basal forebrain, and (iii) the effectiveness of the immunotoxin ME20.4 IgG–saporin for inducing specific and extensive BFC lesions.
Conclusion
The results of this study extend previous work concerning the distribution of BFC neurons in rodents and primates. The general mapping of BFC neurons and their co-localization with p75NTR in sheep is similar to that described previously in mammalian species. Nevertheless, some species differences are present, such as the number of subsectors of NBM cholinergic cell groups. With four subsectors delineated, the organization of the NBM in sheep is intermediate to those of rodents and primates.
