Abstract: Objective: The present study evaluated the endothelial protective effect of the mogrosides extract( ME) from Siraitia grosvenori( Luo Han Guo) in high glucose concentration environments. Siraitia grosvenori,a traditional Chinese medicine,has been shown to exhibit anti-diabetic effects. Firstly,the impairment of endothelium-dependent vasorelaxation in rat isolated mesenteric arteries induced by high glucose was evaluated,and it was observed that pre-incubation of 22. 5m M glucose for 1h significantly impaired the endothelium-dependent vasorelaxation to carbachol. Result: Mogrosides extract( ME,100μg / ml; co-incubated,1h) was able to attenuate the vasorelaxation impairment induced by high glucose( 22. 5m M) and exhibited significant enhancement of the endothelium-dependent vasorelaxation in normal glucose concentration( 5. 5m M). In order to characterize the effects of ME on various endothelium-derived relaxing factors in high glucose environments,blood vessels were co-administered with various inhibitors including indomethacin( 10μM),NG-nitro-L-arginine methyl ester( L-NAME; 100μM)as well as apamin and charybdotoxin in combination( both 100 n M). Conclusion: The results suggested that protective effect of ME against glucose-induced endothelial injuries was partly produced through modulating the production of NO and endothelium-derived hyperpolarizing factor( EDHF) and vasoactive mediators through cyclooxygenase.
