Identification of a Second Murine Interleukin-11 Receptor α-Chain Gene (IL11Ra2) with a Restricted Pattern of Expression

Abstract

The interleukin-11 receptor α-chain, a member of the hematopoietin receptor superfamily, forms, together with gp130, a functional high-affinity receptor complex for interleukin 11. We, and others, reported the cloning of the murine interleukin 11 receptor α-chain cDNA (IL11Ra) and recently described the structure of theIL11Ralocus. We also described the presence of a secondIL11Ra-like locus in some mouse strains. In this study we report that the second locus, designatedIL11Ra2,encodes an mRNA species. The transcript was 99% identical to theIL11Ratranscript in the coding and 3′-untranslated region, but had a different 5′-untranslated region. The complete genomic organization of theIL11Ra2locus is presented, and the two loci are shown to be located on a 200-kbNaeI genomic fragment. Comparison of the expression pattern of theIL11RaandIL11Ra2genes using an RT-PCR restriction fragment length polymorphism strategy revealed that while the expression ofIL11Rawas widespread, expression ofIL11Ra2was restricted to testis, lymph node, and thymus.

    IL-11Rα1, the transmembrane isoform with a short cytoplasmic domain, is found in the brain, bone marrow, muscle, heart, lung, kidney, bladder, spleen, thymus, gastrointestinal tract, salivary glands, testis, ovaries, uterus, macrophages, T-cells, osteoblasts, and osteoclasts [59,60]. IL-11Rα2, the isoform without the cytoplasmic domain, is expressed only in lymph nodes, thymus, and testis [61]. Regulating factors of IL-11Rα expression have not been completely identified [28].

    As shown in Figure 7A, IL-11 and IL-11Rα gene expression was unchanged in gp130Y757F/Y757F thymocytes. Although it has previously been reported that a second IL-11Rα locus (IL-11Rα2) with restricted expression in the testes, thymus, and lymph nodes exists in some strains of mice,8 Q-PCR analyses failed to detect expression of either IL-11Rα locus in thymus and, as expected liver, of gp130Y757F/Y757F:IL-11Rα−/− mice (Figure 7B). Furthermore, unlike gp130Y757F/Y757F mice, IL-11 treatment of gp130Y757F/Y757F:IL-11Rα1−/− mice failed to induce STAT3 tyrosine phosphorylation in lymphoid and nonlymphoid organs (Figure 7C), thus confirming that IL-11 does not elicit a signal in gp130Y757F/Y757F:IL-11Rα1−/− mice.
