Molecular Marker-Based Genetic Analysis of Pasture and Turf Grasses

Abstract

Molecular genetic marker systems have been developed for implementation in the breeding of a number of pasture and turf grass species. A reference genetic map for perennial ryegrass (Lolium perenne L.) has been generated through the activities of the International Lolium Genome Initiative (ILGI), containing conserved restriction fragment length polymorphism (RFLP) loci for comparative genetic analysis with other Poaceae species and simple sequence repeat (SSR) loci for map alignment across multiple pedigrees. Trait-specific genetic mapping in perennial ryegrass and related species is being performed in a number of ILGI-affiliated laboratories, allowing the detection of quantitative trait loci (QTLs) for a range of agronomic traits such as root and shoot morphology, floral development, herbage quality, nutrient assimilation, disease resistance and tolerance to abiotic stresses such as cold and drought. Specific novel strategies are required for effective molecular marker implementation in outbreeding grass species. Candidate gene-based markers provide the means for direct selection of superior alleles in parental genotypes of synthetic varieties and the effective screening and utilisation of large-scale germplasm resources. RFLP and SSR loci corresponding to functionally annotated expressed sequence tags (EST-RFLPs and EST-SSRs) from perennial ryegrass have been assigned to genetic map locations. Corresponding single nucleotide polymorphism markers (EST-SNPs) are under intensive development in our program, with the aim of providing highly informative systems capable of high-throughput automated analysis in breeding programs.
