The Influence of the Intestinal Unstirred Water Layers on the Understanding of the Mechanisms of Lipid Absorption

Abstract

The accurate measurement of the rates of uptake from the intestinal lumen into the enterocyte is central to the elucidation of the cellular processes associated with intestinal absorption of lipids. From the knowledge of the correct rates of uptake, it is possible to determine the permeability properties of the intestinal brush border membrane (BBM), and therefore its lipophilic properties. The permeability of the BBM is subject to adaptation in health and disease (Thomson and Wild, 1997a, 1997b). For example, the uptake of lipids increases with the feeding of a saturated as compared with a polyunsaturated diet, with aging, and with diabetes mellitus. For the treatment of obesity or hyperlipidemia, it may be desirable to develop therapeutic strategies that will reduce the absorption of lipids. Therefore, it becomes necessary to ensure that the experimentally determined rates of lipid uptake are correct, and that the mechanisms of lipid uptake are correctly understood.
