Summary
High doses of N-nitrosomorpholine (NNM) frequently induce cystic cholangiomas in the rat. Three distinct stages can be identified during cholangioma development. In the first stage, as a consequence of a highly dose-dependent toxic necrosis of the liver parenchyma, the bile duct epithelia and mesenchymal cells proliferate; follows fibrosis. In the second stage a focal cholangiofibrosis develops in many animals within weeks or months. At the same time the initially oval or cuboidal cells of the bile-duct epithelia transform into cylindrical cells, which store and secrete abundant mucous substances containing both acidic and neutral mucopolysaccharides (“cholangiolar mucopolysaccharidosis”). In the third stage, reached after a latent period of weeks or months, the foci of cholangiofibrosis give rise to cystic cholangiomas. The cyclindrical mucus-producing epithelium becomes flattened and synthesizes no mucous substances. In some animals the typical pattern of a polycystic disease of the liver is observed. The cholangiolar mucopolysaccharidosis is compared with hepatocellular glycogenosis which precedes the formation of NNM-induced hepatomas. It is suggested that both phenomena are caused by toxic impairement of the cellular carbohydrate metabolism.
