Summary
Some parameters of Au/SH antigen micro CFT were investigated: dependance of maximum titer on complement dosis, incubation time and temperature; kinetics of the complement fixing antigen-antibody-reaction at +4°C; characterisation of the antigen-antibody-reaction in the two-dimensional CFT on the basis of the 50% hemolysis (isofixation curves); comparison of antigens of different concentration by means of the isofixation technique. The highest specific antigen-titers were obtained, when the amount of complement did not exceed 2C′H50, and cold incubation conditions at +4°C were applied. Using an indicator system with low concentration of sensitized sheep cells (o.d. 0.2 at 546 nm) CFT proved to be 200–300fold more sensitive than counter current electrophoresis and agar gel precipitation tests.
There seems to be a correlation between the clinical course of the disease and the amount of cf Au/SH antigen in the patients' serum. Thus, quantitative determination of Au/SH antigen might also be of prognostical value. In case of an acute uncomplicated hepatitis a continuously decreasing titer can be observed, whereas patients suffering from chronic active hepatitis or any other serious basic disease show a continuously high titer.
Zusammenfassung
Einzelne Parameter der Au/SH-Komplementbindungsreaktion, wie Abhängigkeit des Endwertes von der Komplementmenge, von der Inkubationstemperatur, von der Inkubationsdauer, ferner die Spezifität der Au/SH-Kälte-Komplementbindungsreaktion wurden untersucht und die Isofixationskurven des Au/SH-Antigens wurden ermittelt. Geringe Komplementmengen (2C′H50), die Durchführung der Kälte-Komplementbindungsreaktion und die Inkubation von 18 Std ergeben die sensibelsten Reaktionen.
Es konnte gezeigt werden, daß es sich bei der Au/SH-Antigen-Komplementbindungsreaktion um eine für den Nachweis von Australia-Antigen/Antikörper-Reaktionen spezifische Methode handelt. Die Isofixationskurve zeigte die Identität der 3 untersuchten Antigene, ferner den möglicherweise vorhandenen Lipidanteil und schließlich das Auftreten eines Prozonenphänomens im Antigen-Überschußbereich.
Bei dem Vergleich der 3 Methoden erwies sich die Komplementbindungsreaktion als die sensibelste der Nachweismethoden. Sie war etwa 100–150mal sensibler als die Gegenstromelektrophorese und 200–300mal empfindlicher als der Ouchterlony-Diffusions-Test.
Die quantitative Bestimmung des Au/SH-Antigens mit der Komplementbindungsreaktion im Verlaufe einer Hepatitis-Erkrankung ermöglicht u. U. prognostische Aussagen. Bei der unkomplizierten akuten Hepatitis kommt es zum kontinuierlichen Abfall der Serum Au/SH-Antigenkonzentration in Korrelation zur Besserung des klinischen Bildes. Dagegen weisen gleichbleibend hohe Au/SH-Antigenwerte im Serum möglicherweise auf einen prognostisch ungünstigen Krankheitsverlauf hin.
