Summary
A survey on gradient elution TLC is given. At present four processes are known to effect gradients in the mobile phase (“elution gradients”): frontal analysis of a multicomponent solvent (polyzonal TLC), mechanical alteration of solvent composition in the solvent reservoir (solvent gradient), preimpregnation of the adsorbent by vapors of polar or nonpolar substances (vapor, impregnation gradient), and partial removal of mobile phase during chromatography (flux gradient).
Techniques for TLC with antidiagonal solvent gradient and flux gradient are described here for the first time. Flux gradient TLC using a one-component solvent can replace running-through and multiple TLC; using multicomponent solvents a very intense elution gradient can be produced which leads to a nearly stationary chromatographic equilibrium. In this form the method may be considered as a combination of TLC and fractional distillation of the solvent with 100% reflux giving narrow subtrance spots which are nearly independent from the place and the area of sample application.
