Design and bioactivity of Eudragit® encapsulated pH-Sensitive enteric/gastric soluble fluorescent agent

Abstract

Herein, with a view to improving the poor solubility of curcumin and unfavorable optical property of pyrimidine derivatives, the two types of drugs were successfully combined through several simple reactions, and encapsulated with different types of Eudragit® (intestinal / gastric soluble and osmotic). In order to observe the optical properties of the complexes in human body fluid, UV–vis and fluorescence tests were carried out in different pH buffer solutions. The results showed that the maximum emission wavelength of CP-2 was concentrated between 520–600 nm. With the increase of pH value, the maximum emission wavelength of complex CP-2-L shifted red and CP-2-E shifted blue. TEM results showed that the compound CP-2 was evenly wrapped by four kinds of polymer materials, the spherical particle size was uniform, and the particle size was moderate and could be absorbed by cells. Cell imaging results also confirmed this, although only a small part entered the nucleus, most of them were distributed in the cytoplasm. In addition, through molecular docking to simulate the binding between compounds and proteins, the results show that hydrogen bond is the main force. This study aims to provide new ideas for the development of new antitumor fluorescent agents and new gastrointestinal targeted drug delivery.

Highlights

• Two different types of antineoplastic drugs were successfully synthesized and encapsulated with Eudragit®.

• In PBS, pH increased, the maximum emission wavelength of CP-2-L showed a bathochromic-shift and CP-2-E shifted blue.

• TEM showed that compound CP-2 was tightly wrapped, and the spherical particles were uniform in size.

• Cell imaging showed that some complexes could enter the nucleus and most of them were distributed in the cytoplasm.

• Molecular docking shows that hydrogen-bond is the main force.
