Abstract

Bilateral injections of α-methylnoradrenaline into the area of the nucleus tractus solitarii of the brain stem caused a dose-dependent decrease of systemic arterial blood pressure and heart rate of anesthetized rats. The effects were prevented and even reversed by a preceding injection of the α-adrenoceptor blocking agent phentolamine. Pressor doses of angiotensin II and of arginine-vasopressin at the same site failed to decrease blood pressure and heart rate.

The neurochemical and stimulatory effects of putative metabolites of 3,4-methylenedioxyamphetamine and 3,4-methylenedioxymethamphetamine in rats

Rats were injected SC with a dose of 10 mg/kg (as base) of 3,4-methylenedioxyamphetamine (MDA), or 3,4-methylenedioxymethamphetamine (MDMA), 4-hydroxy-3-methoxyamphetamine, α-methyldopamine and α-methylnorepinephrine, metabolites of MDA, and α-methylepinephrine, a putative metabolite of MDMA, twice daily for either 5 or 7 consecutive doses. The rats were killed 24 h after the last injection and monoamines in discrete brain regions were assayed. MDA, MDMA, 4-hydroxy-3-methoxyamphetamine and α-methyldopamine, but not α-methylepinephrine, decreased the concentration of serotonin (5-HT) in the frontal cortex. MDA and MDMA, but not 4-hydroxy-3-methoxyamphetamine, α-methyldopamine and α-methylepinephrine, also decreased the concentration of 5-hydroxyindoleacetic acid (5-HIAA) in the frontal cortexes. In stimulatory studies, MDA and MDMA, but not their metabolites except α-methylepinephrine, which increased activity at 15 and 30 min, increased locomotor activity from 15 to 180 min following the drug administration.
The electrically stimulated release of [<sup>3</sup>H]noradrenaline from nucleus tractus solitarii slices in vitro is modulated via μ-opioid receptors

The electrically stimulated release of [³H]noradrenaline ([³H]NA) from slices of the nucleus tractus solitarii (NTS) from the rat in vitro was inhibited by the α₂-adrenoceptor agonist, clonidine, in a concentration-dependent manner and enhanced by the α₂-adrenoceptor antagonist, yohimbine. Phenylephrine, isoprenaline, carbachol, quinpirole and SKF 38393, all at 10⁻⁶ M, did not affect the stimulus-evoked release of [³H]NA. The opioid peptides, α- and γ-endorphin, did not have a significant effect on the stimulus-evoked release of [³H]NA; however, β-endorphin reduced it in a concentration-dependent manner. [Leu⁵]Enkephalin also reduced [³H]NA release, but higher concentrations were necessary. The selective δ opioid receptor agonists, [D-Pen²,D-Pen⁵]enkephalin (DPDPE) and [D-Ser²(O-tert-butyl),Leu⁵]enkephalyl-Thr⁶ (DSTBULET), as well as the selective κ opioid receptor agonist, U-69593, were not effective. The selective μ opioid receptor agonist, [D-Ala²,NMePhe⁴,Gly-ol⁵]enkephalin (DAGO), concentration dependently reduced the stimulus-evoked release of [³H]NA to the same extent as β-endorphin did. Naloxone, while having no effect on stimulus-evoked [³H]NA release, antagonized the effect of DAGO. These results corroborate that the release of NA from noradrenergic terminals in the NTS region of the medulla oblongata of the rat is modulated via α₂-adrenoceptors and suggest that the release of NA in the NTS in rats is also modulated via μ opioid receptors.
Quantitative autoradiography of α<inf>2</inf> agonist binding sites in the spontaneously hypertensive rat brain

The relative binding of the α₂ agonist [³H]para-aminoclonidine ([³H]PAC) was examined in discrete regions of the spontaneously hypertensive (SHR) and WKY normotensive rat brains by quantitative autoradiography. Substantially less binding was noted in various brain nuclei known to influence cardiovascular function when comparing the SHR to WKY rat brains. These areas included the nucleus of the solitary tract, dorsal motor nucleus of the vagus, locus coeruleus and paraventricular nucleus of the hypothalamus. This difference in binding was also noted in the bed nucleus of the stria terminalis and the nucleus reuniens of the thalamus. Other brain areas which are not believed to be directly involved in central cardiovascular control such as the cerebral cortex, dorsal lateral geniculate and hippocampus exhibited no significant difference in binding between the two strains. Scatchard analysis of [³H]PAC binding indicated that the difference in binding may be due to a selective loss of the high affinity α₂ agonist sites in the SHR. These results indicate that a selective loss of α₂ receptors in discrete brain cardiovascular control regions of the SHR may contribute to the elevated blood pressure seen in this strain.
Endorphins and the hypotensive response to stimulation of alpha-receptors in the brainstem by alpha-methylnoradrenaline

Opioid peptide involvement in the fall in blood pressure resulting from stimulation of alpha-receptors in the brainstem has been investigated in the urethane-anaesthetised rat. Unilateral microinjection of alpha-methylnoradrenaline into the nucleus tractus solitarii (NTS) induced a doserelated fall in blood pressure and heart rate. The depressor response induced by the amine was prevented by pretreatment with naloxone, administered either subcutaneously or directly into the nucleus. Pretreatment with antiserum to beta-endorphin, applied locally, also blocked the depressor response, however a similar dilution of antiserum to met-enkephalin was ineffective in this respect. The local application of phentolamine into the n. tractus solitarii caused an initial fall in both blood pressure and heart rate, and blocked the cardiovascular changes induced by alpha-methylnoradrenaline for at least 90 min. Pretreatment with the alpha-receptor antagonist attenuated the fall in blood pressure produced by microinjection of beta-endorphin. These results suggest that the fall in blood pressure observed after administration of alpha-methylnoradrenaline involves a beta-endorphin-like peptide, a probable site of this interaction being the n. tractus solitarii.
Distribution of α<inf>2</inf> agonist binding sites in the rat and human central nervous system: Analysis of some functional, anatomic correlates of the pharmacologic effects of clonidine and related adrenergic agents

Using [³H]para-aminoclonidine, α₂ adrenergic binding sites have been mapped in the rat and human CNS using in vitro labeling autoradiographic techniques. In both the rat and human thoracic spinal cord, high densities of α₂ binding sites were associated with the substantia gelatinosa and the intermediolateral cell column. In the rat medulla, high binding site density was observed in the medial nucleus of the solitary tract, dorsal motor nucleus of the vagus, raphe pallidus and the substantia gelatinosa of the trigeminal nucleus, while lower levels of specific binding were found in the lateral and ventrolateral medulla. In the human, a similar distribution was observed. However, significantly lower levels of specific binding were seen in the medial nts as opposed to the dmv. In the rat, high levels of specific binding were seen at pontine and midbrain levels in the locus coeruleus, parabrachial nucleus and periaqueductal gray. In the forebrain, several hypothalmic and limbic regions, including the paraventricular and arcuate nuclei of the hypothalamus, the central, medial and basal nuclei of the amygdala, lateral septum and bed nucleus of the stria terminalis and pyriform, entorhinal and insular cortex were labeled. Each of these regions are involved in either modulating autonomic functions directly or integrating somatosensory and/or affective function with autonomic mechanisms. Further, these regions are interrelated by reciprocal connections, and neurons that utilize nor-adrenaline or adrenaline as their neurotransmitter form a vital part of these connections. Thus, these functional, anatomical and neurochemical correlates of the α₂ binding site distribution establish a neurological basis for the complex pharmacological effects of centrally acting α₂ agonists.
Catecholamines and vasopressin in hindbrain nuclei of hypertension prone and resistant rats

Norepinephrine and epinephrine concentrations in the caudal and rostral part of the nucleus tractus solitarii (NTS) and in locus coeruleus (LC) of Sabra hypertension prone (SBH) rats are 2–4-fold higher than in the parent Sabra (SB) strain; SB rats have higher concentrations than the Sabra hypertension resistant (SBN) rats. Dopamine concentrations were higher in SBH as compared to SB and SBN rats only in the caudal NTS. Vasopressin concentrations in the NTS of SBH were 3-fold higher than the levels found in SB or SBN rats. These data suggest that catecholamines and vasopressin in specific brainstem nuclei are involved in either the pathogenesis or central response to hypertension in SBH rats.
