A potent bidirectional promoter from the monocot cereal Eleusine coracana

The cereal inhibitors used in the analysis were finger millet RBI (UniProt accession P01087), corn hageman factor bifunctional (α-amylase-trypsin inhibitor) inhibitor (CHFI) (DQ147269.1), barleypUP23 nonfunctional protein (Lazaro et al., 1988), barley trypsin inhibitor- CHCl3-MeOH soluble (BTI-CMe) (AJ222974), rye trypsin inhibitor (RyeTI) (Lyons et al., 1987) and wheat α-amylase inhibitor (WAI0.28) (Kashlan and Richardson, 1981). Genome walker library was prepared from the genomic DNA of 15 day old finger millet seedlings using the GenomeWalker Universal Kit (Clontech Laboratories Inc.) by digestion with four restriction enzymes separately followed by adaptor ligation.

This suggests that the band with Rf of 0.28 is not homologous to the well known 0.28 monomeric αAI (Kashlan & Richardson, 1981; Silano et al., 1973), but it is rather a comigrating αAI that shares immune epitopes with modern dimeric inhibitors. It is also interesting to note that some of the T.b. accessions not recognized by immunoblotting with the anti-0.28 PAB after SDS–PAGE were, on the contrary, immunodetected after U-PAGE (i.e. lanes 5, 6, 7 and 13).
