Another serum constituent, α2-HS-glycoprotein (#8, also known as fetuin (Brown et al., 1992)) is a liver-derived protein thought to function as an inhibitor of tyrosine kinases (Arnaud and Kalabay, 2002). LOH11CR2A (#15) is another serum glycoprotein, a von Willebrand factor A-containing protein thought to function as a tumor suppressor (Monaco et al., 1997).

    The original study reported that pp63, a liver-secreted phosphoprotein, inhibited the insulin receptor and downstream substrate phosphorylation signaling.63 We identified pp63 as rat fetuin-A on the basis of its cDNA sequence,64 and it was subsequently questioned whether the activity detected in the original study was actually rat fetuin-A or a copurifying protein.65 Using authentic human fetuin-A, we found little inhibition, if any, of the insulin receptor or of downstream substrates.64

    Expression cloning identified a protein member of the fetuin family (α-2-HS-glycoprotein). Referred to as pp63 or tyrosine kinase inhibitor (TKI), this secreted protein is capable of antagonizing insulin signaling (69, 70). Yeast two-hybrid screening identified PTP-1B, a ubiquitously expressed protein-tyrosine phosphatase implicated in down-regulation of several intracellular signaling pathways including insulin receptor phosphorylation and signaling (71,72).
