To assess usability of new indicators for in situ imaging, we loaded them into cultured neurons using a standard AM ester incubation protocol [64–66]. The rate of ACR-1 accumulation (∼0.16 μM/min or ∼0.33 μM/min at room temperature and 37 °C, respectively) is lower than that of fluo-3 (∼0.39 μM/min), fura-2 (0.8 μM/min), and X-rhod-1 (∼3.1 μM/min) (Supplemental Figure 7).
