A simple method to determine the anomeric configuration of sialic acid and its derivatives by 13C-NMR

Abstract

The anomeric configuration of sialic acid and its derivatives could be determined on the basis of the coupling pattern of C-1 in the gated proton-decoupled or selective proton decoupled 13C-NMR spectra; the α anomer gave a doublet C-1 signal while the β gave a singlet.

The α-anomers gave a doublet C-1 signal while the β gave a singlet in their selective proton-decoupled spectra.

    The exclusive α-stereoselectivity was determined by 1H NMR on the basis of well-established empirical rules which define characteristic chemical shifts for H-3eq and H-3ax protons of neuraminyl residue. Indeed, the H3eq proton was found at as a doublet of doublets at δ 2.76 ppm (J3eq,3ax = 12.4 Hz, J3eq,4 = 4.6 Hz), while the H3ax was observed at the typical lower field of δ 1.81 ppm (app t, 12.4 Hz) [44–48]. Moreover, the regioselectivity at C-3′ of the galactoside residue was readily confirmed by the downfield shift of H-3′ from ∼3.5 ppm to ∼4.1 ppm and C-3’ from ∼72.5 ppm to ∼75.1 ppm (Figs. 1 and 2).
