Lactococcal bacteriocins: mode of action and immunity

Abstract

Bacteriocins are antimicrobial peptides produced by bacteria. Some of those synthesized by Lactococcus lactis have been characterized in great detail recently.

The lactococcal bacteriocins are hydrophobic cationic peptides, which form pores in the cytoplasmic membrane of sensitive cells.

      Nisin is one of the oldest antimicrobial peptides and consists of 34 amino acid residues; it was first isolated from Lactococcus lactis by Mattick and Hirsch in 1947 (Mattick and Hirsch, 1947). When nisin is in close proximity to bacteria, it binds tightly to the pyrophosphate of moiety II on the surface of bacteria and subsequently inserts into the cell membrane and forms pores in the bacterial membrane, leading to cell lysis and death (Venema et al., 1995). Thus, nisin is effective against Gram-positive bacteria, such as E. faecalis, Streptococcus mutans, Listeria monocytogenes, and Bacillus cereus (Tong et al., 2014a,b).

      Class I bacteriocins are those containing modified amino acids such as lanthionine (i.e., lantibiotics), Class II is composed of small heat-stable bacteriocins, Class III is composed of large heat-labile bacteriocins, and Class IV is composed of complex protein bacteriocins involving lipid or carbohydrate for activity (Klaenhammer, 1993). The general mechanism for bacteriocin inhibitory activity is that a bacteriocin recognizes either a specific or general receptor molecule on a sensitive cell which allows it to gain a close proximity to the cell membrane to mitigate membrane interaction and pore formation (Venema et al., 1995; Fimland et al., 2005; Hechard and Sahl, 2002). Since LAB are generally recognized as safe (GRAS) for use in foods, bacteriocins produced by LAB have been proposed for use in the biopreservation of food products by inhibiting spoilage microorganisms or pathogens (De Vuyst and Vandamme, 1994; Cleveland et al., 2001).

      Bacteriocins have a narrow host range and are most effective against related bacteria competing for the same resources or ecological niche. Bacteriocins produced by LAB are of most interest to the food industry, because they have potential use as food biopreservatives to control spoilage and pathogenic bacteria (Venema et al., 1995). The main targets of LAB bacteriocins are the cell membrane and cell wall but they can work through many mechanisms to exert an antimicrobial effect (Deegan et al., 2006).
