Synthesis and biological evaluation of 1,4-diazepane derivatives as T-type calcium channel blockers
Graphical abstract
We have synthesized and biologically evaluated 1,4-diazepane derivatives as T-type calcium channel blockers. In this study, we discovered compound 4s, a potential T-type calcium channel blocker with good selectivity over hERG and N-type calcium channels. In addition, it exhibited favorable pharmacokinetic characteristics for further investigation of T-type calcium channel related diseases.

The pyrazole 17 was alkylated with 2-chloro-N-phenylacetamide (18a),43 2-chloro-N-(2-fluorophenyl)acetamide (18b),44 or 2-chloro-N-(3-cyanophenyl)acetamide (18c)45 in the presence of NaH and NaI in anhydrous DMF to yield the target compounds 19a–c and their positional isomers 20a–c in 67–78% and 3–7% yields, respectively. The positional isomers were separated by MPLC, and their structures were confirmed by NOE experiments.
Synthesis, characterization, and X-ray analysis of new N,N′- disubstituted-1,4-diazepanes
2012, Tetrahedron LettersCitation Excerpt :For this reason, different pathways have been reported, for example, reductive cyclization of N-β-hydroxyethyl-N-methyl-1,3-propanediamine,11 from diarylpiperidones via a Schmidt reaction and subsequent reduction,7a aminolysis of aziridines with ω-aminoalcohols and subsequent Fukuyama–Mitsunobu cyclization,12 by microwave-assisted cyclization and LiAlH4 reduction of cyclic lactams13 and by alkylation of the nitrogens of homopiperazine,14 among other methodologies. Using the condensation reaction of aminoalcohols with α-dicarbonyl compounds, our research group has reported the synthesis of piperazines15 (7), bisoxazolidines16 (8), oxazino-oxazines17 (9) and oxadiazepines18 (10) (Fig. 2). In continuation with our studies on condensation reactions, we describe herein a three step synthesis of 1,4-disubstituted diazepanes (13a–f) through the condensation between ethylenediaminealcohols (11a–c) and dicarbonylic compounds followed by stereoselective reduction of the bisbenzoxazolidines to afford N,N′-disubstituted-1,4-dizepanes.
