Species of root-knot nematodes (Meloidogyne spp.) affecting tobacco in north Queensland, including two new host races of M. arenaria

Abstract

Thirty-six tobacco crops in the Mareeba-Dimbulah tobaccogrowing region of north Queensland were sampled and 50% of these were infected by root-knot nematodes. These nematodes were identified by perineal patterns, reactions of standard differential host plants, ß-esterase phenotypes and restriction fragment length polymorphisms in mitochondrial DNA (mtDNA) and in a I.8kb mtDNA polymerase chain reaction (PCR) product. Field populations were found to contain one or more of M. javanica, M. arenaria race 2 and two variants of M. arenaria race 2. It is suggested that these variants be considered as races 2a and 2b of M. arenaria. These new races can be differentiated from M. arenaria race 2 by their reduced reproduction on Bothriochloa insculpta cv. Hatch and Chloris gayana cv. Callide, by ß-esterase phenotype and by RFLPs of mtDNA and an mtDNA PCR product. They can be distinguished from each other by differential reproduction on Brachiaria humidicola and C. gayana cv. Samford.
