2012, Molecular and Cellular ProbesCitation Excerpt :Because PCR (particularly nested-PCR) is highly sensitive [38,39], there is the potential to detect pathogen DNA in human faeces in the absence of clinical signs of disease, thus the amount of pathogen present also must be considered. Recent studies have demonstrated the utility of MT-PCR for accurate quantification (correlation co-efficient: >0.9) by employing an internal control of known concentration [21,40]. The internal positive spike-control (calculated to be 10,000 copies of a synthetic oligonucleotide) reaction for each sample allowed any inhibition to PCR to be detected and the amount of each specific amplicon to be estimated [21].
